Instructions for use
1. Open the culture tube and stand
it upright. Using forceps take one
capillary from the container (if
possible fl ame the jaws of the for-
ceps before use). Caps should be
placed on the table wide end down
as illustrated.
2. a) Dip the capillary into a tank, or
b) expose the capillary to a spray
or running fl uid, or
c) mix the sample in the container
and dip the capillary fi lling it
totally.
3. Press the free end of capillary gen-
tly against a clean absorbant paper
in order to empty the capillary.
4. Insert the capillary into the cul-
ture medium (so that the capillary
touches the bottom of the culture
tube). Leave the capillary there.
N.B. Handle the capillary only
with forceps. Do not touch it
with your hands or clothing and
do not lay it down. Use a fresh
capillary for each sample.
5. Close the culture tube fi rmly, fi ll
in the label and affi x it to the tube.
Incubate the tube at about 35°C
(95°F).

Interpretation of results
The growth of anaerobic sulfi de gen-
erating bacteria in the Easicult S tube
results in the formation of black iron
sulfi de. The blackening may begin at
any point of the tube and depending
on the degree of contaminaiton even-
tually part of or the whole tube may
become black.
The result is not quantitative but shows
the approximate level of contamination.
Once sulfi de generation has com-
menced it will almost always increase
fairly rapidly. Sulfi de generation can
be prevented by adequate aeration
although
will continue to fl ourish. Suppression
of aerobic micro-organisms will usu-
ally suppress also sulfi de generating
organisms as the former are oxygen
scavengers and create the oxygen
free conditions needed by sulfi de gen-
erating bacteria.
Storage
Easicult S tubes may be stored at
room temperature (abt. 20°C/68°F)
protected from light and draught.
The expiry date is marked on the box.
If there should be any visible growth
of bacteria or fungi in an unused tube,
it should be discarded.
Disposal of used tubes
As the incubated tubes are bacterial
cultures, they should be handled care-
fully. Disposing of used tubes can best
be done by burning or by autoclaving
them after loosening the caps (a pres-
sure cooker can be used for this).
