Kinetex
2.6 µm core-shell columns operate comfortably within
®
the pressure limits of conventional HPLC instruments and rival the
performance of sub-2 µm fully porous particles achieved on UHPLC
instruments. However, to maximize the benefits from your Kinetex
core-shell column do the following:
•
Minimize sample dispersion before the column
1. Gradient elution will minimize dispersion
2. In isocratic elution, use an injection solvent that's weaker than
your mobile phase
•
Optimize detector settings by adjusting the scan rate and/or the time
constant to the fastest possible setting such that signal-to-noise
(s/n) is not adversely affected.
•
Minimize the extra-column volume between the injector and the
column, and between the column and the detector
1. Minimize the length of all connection tubing
2. Use 0.12 mm ID (0.005 in.) tubing whenever possible
a) 0.17 mm ID (0.007 in.) tubing is acceptable
b) Avoid, if possible, using 0.25 mm ID (0.010 in.) tubing
3. Use extremely low dead-volume fittings
4. Ensure all connecting tubing is seated properly at every
connection
•
Use a micro volume flow-cell
1. Standard flow cells on conventional LC systems can be >10 µL
2. For best results, replace standard flow cells with <3 µL flow cells
(<2 µL when using 2.1 mm ID columns)

Extending Column Lifetime
Phenomenex recommends the use of the SecurityGuard
guard cartridge system to extend the lifetime of your Kinetex column,
especially with samples extracted from complex matrixes. Ideally,
samples must be completely dissolved in the mobile phase or filtered
through a syringe filter of approximately 0.20 µm porosity.
