5.2 Performing Diafiltration
Material: — 5x sample volumes of exchange buffer
— Diafiltration tubing
— Exchange buffer vessel
Requirements
The sample volume is ≤ 500 mL.
WARNING
Risk of infection due to leakage resulting in contact with solutions!
During the process, check the system for leaks.

Observe the operating conditions (chapter "6.4 Operating Conditions",

page 22).
Procedure
Empty the permeate vessel.

Fill the exchange buffer vessel, e.g. laboratory bottle, with 5x sample vol-
t
umes of an appropriate exchange buffer.
Connect exchange buffer tubing to the exchange port on the feed reser-
t
voir and place the other end to the bottom of the exchange buffer vessel.
Start the peristaltic pump with a suitable flow rate (see chapter "6.4 Oper-
t
ating Conditions", page 22).
The diafiltration process starts.

The pressure indicator should read up to 2.5 bar.

When the required volume of exchange buffer has been added to the sys-
t
tem and the desired retentate volume has been reached: Stop the peri-
staltic pump.
The diafiltration process is complete.
