Low purity of target cells
A low purity of target cells in sort can be caused by:
Dim staining.
l
Event and/or sort rate is too high.
l
Low frequency of target cells.
l
Valve timing and/or opening time is too long.
l
Do one or all of the following:
Change antibody conjugate or use additional marker.
l
Adjust sample concentration.
l
Immunomagnetic pre-enrichment or two-step sort.
l
Reduce max. opening time in valve table programmer (refer to MACSQuantify Tyto Software User manual).
l
