Freshly collected serum or urine samples are the specimens of choice. Whilst serum samples can be
stored at 15...30°C for up to 4 days, 2...6°C for up to 2 weeks or 6 months at -20°C for standard 5

band patterns
, the beta 2-globulin band (Complement C3) will degrade rapidly on storage. Urine
samples can be stored refrigerated at 2...6°C for up to 72 hours or 2 weeks at -20°C. Urine samples
should initially be used without concentration or dilution, unless the total protein is known to exceed
1000mg/L.

1. URINE: Pipette 35µl of sample into the appropriate disposable sample cups. Protect the samples
from evaporation.
SERUM: Dilute each sample to a 1 + 2 dilution, using IFE diluent. Pipette 35µl of diluted sample
into the appropriate disposable sample cups. Protect the samples from evaporation.
2. Switch electrophoresis unit on and:
i i ) )
S S A A S S - - 1 1 P P l l u u s s u u s s e e r r s s : : Carefully place the sample tray onto the applicator drawer. Ensure that the tray
is pushed firmly down into position.
i i i i ) ) S S A A S S - - 3 3 u u s s e e r r s s : : Carefully locate the sample tray using the sample base locating pins. Ensure that the
tray is positioned securely.
3. Remove the gel from the packaging and:
i i ) )
S S A A S S - - 1 1 P P l l u u s s u u s s e e r r s s : : dispense 400µL of REP Prep onto the heat sink. Place the gel onto the heat sink,
agarose side up, aligning the positive and negative sides with the corresponding electrode posts,
taking care to avoid air bubbles under the gel.
i i i i ) ) S S A A S S - - 3 3 u u s s e e r r s s : : place the alignment guide onto the pins and dispense 400µL of REP Prep onto the
centre of the chamber. Place the gel into the chamber agarose side up, using the guide, align the
positive and negative sides with the corresponding electrode posts, taking care to avoid air bubbles
under the gel.
4. Blot the gel surface with a blotter C, discard the blotter.
5. i i ) ) S S A A S S - - 1 1 P P l l u u s s u u s s e e r r s s : : Place the cover over the gel and electrodes and press firmly for 5 seconds to
ensure contact.
i i i i ) ) S S A A S S - - 3 3 u u s s e e r r s s : : attach the electrodes onto the the electrode posts so that they are in contact with
the gel blocks.
6. Place 2 applicator blade assemblies in position on the instrument, ( ( S S A A S S - - 3 3 u u s s e e r r s s : : s s l l o o t t A A a a n n d d 1 1 0 0 ) ) .
7. Perform the Pentavalent Screen electrophoresis:
i i ) )
S S A A S S - - 1 1 P P l l u u s s u u s s e e r r s s : : Electrophoresis: 100 volts, 18 mins, 20°C
N N O O T T E E : : For Serum Screen, 1 sample application is required. For Urine Screen, 10
sample applications are required.
Incubation Step 1: 8 mins, 37°C (incubate)
Incubation Step 2: 8 mins, 40°C (D blot)
